Parkinson's Disease Review Articles

Parkinson's disease (PD) is a complex neurological disease, affecting approximately 2% of the population over the age of 60. Since the first reports of PD correlation with the SNCA gene, an increasing number of genomic predispositions have been identified as direct contributors or risk factors for the disease. Over the past decade, advances in cell reprogramming and induced pluripotent stem cell (iPSC) technology have led to the development of brain tissue engineering derived from patients. The cultivation of living neurons derived from patients offers a unique opportunity to study in vitro the cellular mechanisms of genetically linked diseases. IPSC studies of brain disorders remain laborious and expensive, which limits the number of cell lines that can be studied in a single laboratory and can pose the problem of statistical power and reproducibility. The field is still in its infancy and neuronal differentiation protocols are continuously improved, which makes technical harmonization between studies difficult. Like any model, iPSC studies have their weaknesses and the current limits are tackled collectively by the research community. However, the urgent need for better treatments for brain disorders justifies the need for pioneering iPSC studies modeling these diseases in vitro now. Since 2011, more than 385 neural lines of PD patients have been generated through 67 independent originals.
