Abstract :

Sortase A (SrtA) is an enzyme from the transpeptidase family and plays a key role in the adhesion to and invasion of hosts by Gram-positive bacteria. Streptococcus mutans is deemed to be the major pathogen responsible for the generation of dental caries. Since SrtA anchored proteins contribute to the adherence of S. mutans and srtA knockout, S. mutans variants showed a decreased ability to colonize the oral mucosa and teeth and consequently reduced caries formation. Therefore, this enzyme could be a future target for the prevention of dental caries. We found that the organic extract from the dried flowers of Sophora japonica significantly inhibited SrtA derived from S. mutans isolated from the human oral cavity. Using repetitive column chromatography and HPLC, seven compounds were isolated from dried flowers of S. japonica. On the basis of spectroscopic analysis, these compounds were identified as quercetin, two quercetin derivatives and four maltol derivatives. These compounds inhibited the action of SrtA from S. mutans without affecting cell viability. The onset and magnitude of inhibition of salivainduced aggregation of S. mutans treated with compound was comparable to that of untreated S. mutans with a deletion of the srtA gene.
