Tauopathies are characterized by the aberrant modification, splicing and aggregation of the Tau protein associated with microtubules into filamentous inclusions in neurons and glia. Tau pathology is present in a wide range of neurodegenerative diseases, including Alzheimer's disease, frontotemporal dementias, Pick's disease, progressive supranuclear palsy, chronic traumatic encephalopathy, Huntington's disease and Parkinson's disease, among others. Given the combined prevalence of these diseases and the importance of Tau pathology in Alzheimer's disease, research on tauopathy should be considered a priority.

Although great progress has been made so far, a number of unresolved questions remain regarding the molecular and cellular mechanisms by which Tau pathology (in the presence or absence of tau mutations) is initiated, amplified and propagated ; and how it evolves towards the different characteristics of tauopathy leading to dementia and shrinking of the brain. In addition, although a number of studies have indicated that early cognitive deficits are caused by synaptic dysfunction and neuritic connectivity deficits, the underlying mechanisms and how they cause clinical symptoms are not entirely included; a subject of great relevance in the human context.
