The nucleotide sequence of the gene was established. There are variety of isolation protocols but all require digestion, decontamination, and concentration steps. The mycobacteria must be recovered undamaged while organic debris is liquefied and contaminating microorganisms are killed. The cell membrane may be a major virulence factor of tubercle bacillus and contributes to its intrinsic drug resistance. Recently, cryo-electron microscopy showed that mycobacterial cell membrane lipids form an unusual outer membrane. Meanwhile, nitric oxide (NO) is a member of reactive nitrogen species (RNS) generally considered to play a key role in the bactericidal process in innate immunity against Mycobacterium tuberculosis complex infection. The present study therefore investigated the mechanism of NO production in murine DCs induced by Mycobacterium bovis (M.bovis) and its attenuated strain Bacillus Calmette-Guérin (BCG) infection. While the precursors could not be isolated directly due to their inherent instability during base treatment, their presence was established by prior reduction of the β-oxo group by sodium borohydride. Interestingly, the mutant also accumulated unsaturated ketones, almost like tuberculenone from M. tuberculosis, which were shunt products derived from spontaneous decarboxylation of α-alkyl, β-oxo carboxylic acid precursors of mycolic acids.
