The smallest member of the apoptosis-inhibiting protein (IAP) family, involved in the inhibition of apoptosis and the regulation of the cell cycle. These functional attributes make Survivin a unique protein with divergent functions, that is to say, regulating cell proliferation and cell death. Survivin's pattern of expression is also distinctive; it is expressed prominently during embryonic development, absent in most normal terminal differentiated tissues but upregulated in a variety of human cancers. The expression of survivin in tumors is correlated not only with the inhibition of apoptosis and a decrease in the rate of cell death but also with the resistance to chemotherapy and the aggressiveness of tumors. Therefore, Survivin is an important target for cancer vaccines and therapies. Survivin has also been found to be prominently expressed in both human and embryonic stem cells and in many types of somatic stem cells, indicating its as yet unexplored role in the generation and maintenance of stem cells. Overall, Survivin is emerging as a molecule with a much broader role in cell homeostasis. This review will discuss various aspects of the biology of survivin and its role in the regulation of apoptosis, cell division, chemoresistance, and tumor progression. Various molecular and immunotherapeutic approaches targeting survivin will also be discussed.
