Complementary rat DNA (cDNA) for the beta RI isoform of the adenosine monophosphate dependent protein kinase I (cAMP) regulatory subunit has been cloned and sequenced and found to be contained all of the protein coding and the 3 'untranslated regions, with a single (ATTAAA) polyadenylation site. The larger open reading frame was preceded by a short phase shifted open reading frame, which is not seen in the corresponding mouse RI beta cDNA due to a single basic substitution. The rat RI beta cDNA clone was 2374 bases in length and detected a rat mRNA of about 2.8 kilobases. Rat RI beta mRNA was abundant in the adult rat brain and testes, but was not detectable in other rat tissues. Rat beta RI cDNA also detected beta RI mRNA in mouse brains, but not mouse testes, from 10 week BALB / c or 10 and 6 week old Swiss Webster mice. Thus, despite a 96% nucleotide identity in the RI beta coding region in rats compared to mice, there are at least two differences in these closely related species. First, there is a short open reading frame, which precedes the coding region in the rat but not in the mouse.
