Determination of Pregabalin in Tissues and Fluids by Using GC-MS

Pregabalin is an antiepileptic and analgesic drug, commercialized under the name of Lyrica and other names, generally used to treat neuropathic pain. The determination of pregabalin was quantified in tissues and fluids samples by using Gas chromatography coupled with mass spectrometry, isolation and precipitation protein by Ammonium Sulphate method. The limit of detection (LOD) is 200 ng and the limit of quantification (LOQ) in is 400 ng. Ibuprofen was used as internal standard, this methods has been applied in two cases of pregabalin.

Identification of Metabolites using 1H NMR in Polycystic Ovary Syndrome Cases from North India

Polycystic Ovary Syndrome (PCOS) is a common, heterogenous endocrine disorder in women of reproductive age. Alterations in numerous metabolic pathways have been implicated in its pathophysiology. The systemic changes in PCOS initate not only changes in the ovarian function but also changes in the whole-body metabolism. Indian women are at higher risk of developing type 2 diabetes and insulin resistance and hence give the drive to carry metabolomics study on Indian women. The present study focused on the metabolic profiling of PCOS women in North India. A total of 28 females of reproductive age were included. 1HNMR technique was used to identify metabolites. Orthogonal projection to the latent structure with discriminant analysis (OPLS-DA) was applied for model identification. Important metabolites were identified using variance of importance (VIP) score and s-plot and the significance was verified by univariate analysis. The levels of leucine, glutamine, citric acid, proline and, methionine were decreased and that of valine, tyrosine and phenylalanine were elevated in PCOS women as compared to control females. The differences in the metabolic profiles between PCOS cases and controls may be attributable to the alterations in various pathways. The present study is first of its kind from North India.

Isolation, Characterization and Quantification of Civetone from Civet Musk

Civet is an important commodity of commerce because of its use in perfume industry. It is also used to some extent in traditional medicine. Civetone is the main component of civet that is responsible for its characteristic odor. The amount of civetone in the musk of civet cat is not determined yet. In this project, rapid, simple and sensitive quantitative thin-layer chromatography method for the quantification of civetone in civet was developed and validated. The method involved extracting samples with dichloromethane by sonication, and analysis by CAMAG TLC scanner 3, after using a civetone dervatizing reagent. The densitometric scanning and analysis was done in absorbance mode at 371 nm. A new method for quantifying civetone in civet was developed in the course of this study. This involved taking a known amount of civet adding to it, a dervatizing agent 2,4-dintrophenylhydrazine, which now raises the absorption maximum to the visible range, thus making quantification work possible. The linear regression analysis data for calibration plots showed good linear relationship with r2=0.999 in the concentration range of 1-5 μg spot-1 with respect to peak area. The method was validated for accuracy, precision and recovery. The civetone value in crude civet samples, quantified by TLC was 0.8-1.2%.

An Efficient One Pot Synthesis and Biological Screening of Some Pyrazole Based Heterocycles

Present work deals with the preparation of some Pyrazole derivative salts which was prepared by using different kinds of aldehydes and hydrazine derivatives by using alkoxide as a base and using the 1,3- dicarbonyl compound like ethyl acetoacetate at room temperature. The reaction mixture was stirred on magnetic stirrer for about 10 to 20 minutes till colour change is observed and then product is separated by using organic solvent. The product obtained was converted to sodium salt of Pyrazole derivative by base hydrolysis. The entire synthesized compound was tested for their antibacterial activity against the Gram-positive and Gram-negative strains of bacteria. The investigation of antibacterial screening data revealed that most of the tested compounds showed moderate to good antibacterial activity.
